One known example
I know that there are a few sets that will be difficult to complete because there are only one or two examples of a particular card known to exist. The Wagner throwing from the e105 Mello Mint set is one that comes to mind. What other individual cards makes collecting a particular set near impossible?

Re: One known example
I think E107's would fall into this group. I don't know about 1 of 1's, but there are definitely many cards where no more than 5 are known to exist.
I love this set, but due to the sheer value of these cards, it would be near impossible, unless I had endless pockets....which I don't.

Another set that comes to mind is the T216 set. Especially if you try to collect the harder to find Mino and Virginia Extra backs. Many of the Virginia Extra's are 1 of 1 or maybe 1 of 2. For others, there aren't even any known examples.

e104-3s have several 1 of 1 players in the set. If you want to get into whether it has a Nadja back or not, then you have even more 1 of 1 cards in the set.
Zeenuts also have some 1 of 1 cards. Like OJs, the true rarities of the set are jealously gaurded secrets.

It would be had to collect N167's. They can cost thousands for a common. There are only 12 cards in the set though.

Re: One known example
There are a number of N173s that have but a single example known....not that there are more than a small handful of people that even think of that in terms of a "set"

The same can be said about the w600 set. The checklist is massive, but many were probably never produced because of the lack of player's popularity. Many others had only a few made, meaning that the chance of surviving to this day is slim.

Alphas, Just So Burkett, lots of Negro Leaguers including Pete Hill and Willie Foster. There are many uncataloged cards.

Re: One known example
Out of all the backs of the m101-4/5's . only about 3 backs could be completed. Sporting news, Blank, Famous & Barr Maybe but it would be a tough go.
Re: One known example
All 104 Allegheny cards are unique as are the T231 Fans cards (or card?) Also the Maple Crisp Stengel and Holland Creameries Peckinpaugh have only one known example, while the US Carmel Lindstrom has two. Plenty of obscure sets have cards where there may only be one example: Leader Novelty, Schapira Candy, J=K, All Star Baseball, Tango Eggs, and T4 Obak, to name a few. Plenty of backs that are unique: T207 Red Cross, E92 Croft's Red back, T216 Virginia Brights, and probably a few T205/6 Drum and Hindus.

one of a kind cards
There are several one of a kind cards in the E125 set. Also the T214 Victory set. Maybe T215 Pirates and T216 Virginia Extra as others have already mentioned. Is there more than one E92 Nadja Cobb? Quan - I think there is at least one other mello mint throwing wagner on top of your list.
